qnrB, another plasmid-mediated gene for quinolone resistance.

چکیده
A novel plasmid-mediated quinolone resistance gene, qnrB, has been discovered in a plasmid encoding the CTX-M-15 beta-lactamase from a Klebsiella pneumoniae strain isolated in South India. It has less than 40% amino acid identity with the original qnr (now qnrA) gene or with the recently described qnrS but, like them, codes for a protein belonging to the pentapeptide repeat family. Strains with qnrB demonstrated low-level resistance to all quinolones tested. The gene has been cloned in an expression vector attaching a polyhistidine tag, which facilitated purification to >or=95% homogeneity. As little as 5 pM of QnrB-His6 protected purified DNA gyrase against inhibition by 2 microg/ml (6 microM) ciprofloxacin. With a PCR assay qnrB has been detected in Citrobacter koseri, Enterobacter cloacae, and Escherichia coli isolates from the United States, linked to SHV-12 beta-lactamase and coding for a product differing in five amino acids from the Indian (now QnrB1) variety. The qnrB gene has been found near Orf1005 in some, but not all, plasmids and in association with open reading frames matching known chromosomal genes, suggesting that it too was acquired by plasmids from an as-yet-unknown bacterial source.

The Emergence of Qnr-resistance Among Klebsiella pneumoniae spp in Zahedan
Objective: Although quinolone resistance results mostly from chromosomal mutations in Klebsiella pneumoniae, it may also be mediated by plasmid – encoded qnr determinants. Plasmid-mediated quinolone resistance (PMQR) was increasingly identified in Enterobacteriaceae family worldwide. The aim of this study was to investigate the prevalence of qnr genes in clinical isolates of Klebsiella pn...
متن کاملPrevalence of Plasmid-Mediated Quinolones Resistance among Klebsiella pneumoniae Strains Isolated from Hospitals in Borujerd, Iran
ABSTRACT Background and Objective: Klebsiella pneumoniae is one of the most common causes of bacterial infections. Presence of plasmid-mediated quinolone resistance genes causes low level of resistance in K. pneumoniae. This study investigated the prevalence of resistance to quinolones and fluoroquinolones, and the frequency of qnrA, qnrB and qnrS genes amo...

Objective(s): Plasmid-mediated quinolone resistance (PMQR) determinants and integrons have a considerable contribution to bacterial drug resistance in Gram-negative pathogens. We studied the prevalence of PMQR genes and integron carriage in multidrug-resistant community isolates of Klebsiella spp.Materials and Methods: Two hundred and fi...
متن کاملPrevalence of plasmid-mediated quinolone resistance determinants among oxyiminocephalosporin-resistant Enterobacteriaceae in Argentina
High quinolone resistance rates were observed among oxyiminocephalosporin-resistant enterobacteria. In the present study, we searched for the prevalence of plasmid-mediated quinolone resistance (PMQR) genes within the 55 oxyiminocephalosporin-resistant enterobacteria collected in a previous survey. The main PMQR determinants were aac(6')-Ib-cr and qnrB, which had prevalence rates of 42.4% and 3...
متن کاملThe main purpose of this study was the pattern of antibiotic resistance and the frequency of plasmid-dependent quinolone resistance (qnr) genes in Pseudomonas aeruginosa, Escherichia coli and Klebsiella pneumoniae isolates isolated from surgical site
Introduction: In recent years, antibiotic resistance has been on the rise, leading to limited ways to control nosocomial infections (especially surgical site infections) and the right treatment options. The main purpose of this study was the pattern of antibiotic resistance and the frequency of plasmid-dependent quinolone resistance (qnr) genes in Pseudomonas aeruginosa, Escherichia coli and Kl...
